TAAB社包埋用樹脂

Epon 812 is the triglycidyl ether of glycerol and was developed in the USA as an alternative to Araldite CY212. It is used in a combination with DDSA and methyl nadic anhydride (MNA) and is rather less viscous than Araldite mixtures. A suitable mixture for medium hardness blocks is:

Epon 812 48.0 parts DDSA 19.0 parts MNA 33.0 parts DMP-30 2.0 parts

Harder blocks may be obtained by replacing part of the DDSA by MNA and softer blocks by replacing part of the MNA with DDSA. Some experimentation is worthwhile to achieve the perfect formulation for the tissue most commonly sectioned.

パラプラストプラス PARAPLAST PLUS 502004 (Tissue Embedding Medium by McCormick Scientific )

A compound of purified paraffin and plastic polymers of regulated molecular weights. Contains Dimethyl sulphoxide (DMSO) for rapid tissue infiltration.

Paraplast Plus tissue embedding media is a mixture of highly purified paraffin containing a plastic polymer. Paraplast Plus contains a small percentage(0.8%) of dimethyl sulphoxide(DMSO). The plastic polymer combined with paraffin yields a media whose physical properties enhance quality and ease of sectioning. DMSO in paraplast Plus further enhances these properties.

Background and Advantages: The introduction of Spurr resin provided the microscopist with a resin of exceptional penetration qualities. Unlike the other epoxies, Spurr resin with a viscosity of 60 cps will readily penetrate membrane walls, hard tissue, rocks and other minerals. It has also been used successfully in preparations of specimens with a high lipid content, tissues with hard, lignified walls, and highly vacuolated parenchymatous tissue.

The Spurr resin is based on the four following components. ERL 4221, which because of its compact structure yields highly cross-linked polymers with good high temperature resistance.

The flexibilizer is the epoxy resin, D.E.R.736,(diglycidyl ether of polypropylene glycol). This has a WPE value of 175-205. The amount of D.E.R. 736 may be regulated to provide embedments with a variety of hardness characteristics.

NSA, Nonenyl succinic anhydride is the hardener specially purified for EM usage. To avoid hydorlysis of epoxide or anhydide bond, we recommend minimum exposure of the above to atmospheric moisture. The accelerator is demethylaminoethanol(DMAE). This agent gives longer pot life the resin with a lighter color. Curing times can be shortened by increasing the accelerator ratios.

Lowicryl Embedding Media

Tissue embedded in JB-4 Embedding Medium offers superior ultrastructural preservation and semi-thin sections for high resolution light microscopy. This water solubleplastic media can replace most embedding procedures in which pafaffin or epoxy resins are used, and offers the advantage of less distortion and tissue shrinkage. Difficult specimesn, such as undecalcified bone and delicate embryonic tissue can be easily processed in JB-4 plastic.

Both aqueous and non-aqueous fixatives can be utilized with JB-4 embedding medium including OsO4 and neutral buffered fixatives. Unlike paraffin, removal of JB-4 plastic prior to staining is unnecessary and brilliantly clear saining has been achieved using water soluble dyes. JB-4 does not interact with any tissue group pf importance for staining during polymerization, contrast to epoxy resins which may alter the staining character of tissue and are less responsive to histological stains.

Extremely fast processing times are possible with JB-4 embedding medium as only a few simple processing steps are required. Since JB-4 is water soluble, there is no need to completely dehydrate the tissue before infiltrating. A graded series of ethyl alcohols through 95% is commonly used to dehydrate tissue. For the retention of lipids and enzymes, precessing tissue at 4º C is recommended.

On a routine basis, 1-2µ thick sections are easily cut with a glass knife. Recently some investigators have even utilized conventional rotary microtomes for semi-thin sections of JB-4 embedded material. Under the same conditions, paraffin sections must be cut 5-6µ thick with much cellular overlap and the loss of clear cellular resolution. Traditional hard embedding plastics, such as epoxies or polyesters are more difficult to sections at the 1-2µ level and often result in wrinkling and compression.

JB-4 has been used sucessfully in enzyme histochemistry, immunohistochemistry techniques, and high resolution autoradiography sections, JB-4 plastic enhances the ability to identify labelled cells.

The increased unclear differentiation obtained with JB-4 plastic is a great aid to diagnosis, especially with liver and kidney tissue where fine cytogogical detail and rapid processing is necessary. Such morphological evaluation from needle biopsies is not possible with paraffin or other plastic materials.

血管注入剤 Microfil の素材はシリコンラバーで、樹脂状の血管注入剤とは異なり、決して硬化はせず、注入した臓器からも通常のパラフィン包埋組織標本を作製する事ができます。 Microfil の最も優れている点は、微細な毛細血管レベルまで良好な血管網が描出される点で、通常のバリウムゼラチン法とは比較にならないほどの美しさです。

  1. 極めて微小なレベルまで良好な充溢感が得られ、従来のバリウムゼラチン法をはるかに凌駕しています。

  2. 簡単にブレンドが得られます。

  3. 透徹法による立体観察の外、通常の組織検査にも用いられ、また軟X線撮影も可能です。

MICROFIL compounds will fill and opacify microvascular and other spaces of non-surviving animals and post-mortem tissue under physiological injection pressure. The continuous, closed vascular system lends itself to flow through or perfusion techniques. Following injection, MICROFIL compounds cure to form a three-dimensional cast of the vasculature.

MICROFIL MV-series compounds are available in five radiopaque colors, as well as clear. MV-series compounds require either an alcohol-methyl salicylate or glycerin clearing sequence, whereby the refractive index of the clearing solution is the same as the refractive index of the tissue. This allows for microscopic examination of a selected vasculare bed.

MICROFIL CP-101 compound is intended for use in cast-corrosion techniques, and is designed for filling large blood vessels(greater than 100microns). Although the CP-101 compound will fill capillaries, these vessels fragment when the supporting tissue is removed through exposure to a potassium hydroxide solution. When cured, MICROFIL CP-101 is milky white in color, Casts made using CP-101 will maintain their dimensional accuracy indefinitely.
